Overview

Edicotinib is an orally active, brain penetrant, small molecule inhibitor of the macrophage colony-stimulating factor 1 receptor (CSF1R), also known as FMS or CD115. It selectively blocks CSF1R tyrosine kinase activity, thereby inhibiting the survival, proliferation, and differentiation of macrophages and microglia. This mechanism is relevant in diseases where macrophage-driven inflammation or microglial activation play a pathogenic role. Edicotinib has been investigated for use in autoimmune diseases such as rheumatoid arthritis and Crohn’s disease, neurodegenerative conditions like Alzheimer’s disease, and various cancers including acute myeloid leukemia and Hodgkin's lymphoma. While it showed safety in clinical trials, efficacy endpoints were not met in several indications leading to discontinuation for some diseases[2][3][5][6].
