Edonerpic is an orally available small molecule neurotrophic agent, specifically a benzothiophenyl derivative, developed primarily for the treatment of Alzheimer's disease and stroke. It is being developed by FUJIFILM Toyama Chemical (formerly Toyama Chemical). Edonerpic acts as a neuroprotectant and has been shown to enhance cortical synaptic plasticity and motor function recovery after brain injury or stroke in animal models. Its mechanism of action involves binding to collapsin response mediator protein 2 (CRMP2), which regulates neurite outgrowth, ion channel trafficking, and synaptic plasticity; this interaction promotes AMPA receptor trafficking to the postsynaptic membrane, thereby enhancing neural network reorganization and functional recovery. There is also evidence that it modulates both AMPAR and GABAR-mediated currents in compensatory brain regions during recovery from neurological damage[1][4][7][8]. Clinical trials have focused on Alzheimer's disease (Phase II) and stroke (Phase II), but results so far have not demonstrated significant efficacy in altering key biomarkers or clinical outcomes in Alzheimer's disease[1][4].
