edTreg (engineered dominant Regulatory T cells) is a proprietary cell therapy platform developed by GentiBio to treat autoimmune and inflammatory diseases. The technology involves the conversion of abundant autologous or allogeneic effector T cells (Teffs) into stable, functional regulatory T cells (Tregs) through the lentiviral transduction of a FOXP3 transgene. Unlike natural Tregs, which are rare and can be phenotypically unstable in inflammatory environments, edTregs are designed to maintain their suppressive identity. They are further engineered with a Chimeric Antigen Receptor (CAR) or T-cell receptor (TCR) for tissue-specific targeting and a Rapamycin-Activated Cytokine Receptor (RACR). The RACR system allows for the selective expansion and persistence of the edTregs in vivo using low-dose rapamycin as a survival signal. The lead candidate, GNTB-120, is currently in clinical development for the treatment of Type 1 Diabetes, aiming to restore immune tolerance and protect pancreatic beta cells from autoimmune destruction.
