EDV-ANDC is a first-in-class Antibody Nanocell Drug Conjugate (ANDC) being developed by EnGeneIC. The therapeutic platform utilizes EnGeneIC Dream Vector (EDV) nanocells—non-living, bacterial-derived nanoparticles—that are loaded with a potent cytotoxic payload. These nanocells are targeted to cancer cells using bispecific antibodies, which bind to both the nanocell surface and a specific tumor cell receptor (typically EGFR in EnGeneIC's lead programs). Upon binding, the nanocells are internalized via endocytosis, releasing the cytotoxic payload intracellularly. Simultaneously, the bacterial components of the nanocell membrane act as an adjuvant, stimulating the patient's innate immune system and promoting a systemic anti-tumor immune response. This dual 'cyto-immuno' mechanism is designed to enhance efficacy and overcome drug resistance in intractable cancers. EDV-ANDC has received FDA Fast-Track designation for the treatment of pancreatic ductal adenocarcinoma (PDAC).
