eEF-2K siRNA is a research-stage small interfering RNA (siRNA) therapeutic designed to silence the expression of eukaryotic elongation factor 2 kinase (eEF2K). eEF2K is a calcium/calmodulin-dependent serine/threonine kinase that is frequently overexpressed in various malignancies, including triple-negative breast cancer (TNBC), pancreatic cancer, glioblastoma, and melanoma. By targeting eEF2K mRNA for degradation, the siRNA inhibits the phosphorylation of eEF2, thereby disrupting protein synthesis elongation and inducing apoptosis or autophagy-mediated cell death. Preclinical studies have utilized neutral nanoliposomes (such as DOPC) for systemic delivery, demonstrating significant antitumor activity and sensitization to chemotherapy.
