eExos-miR-135 is an engineered exosome-based gene delivery system designed for the treatment of glioblastoma. These exosomes (eExos) are modified to express retroviral Gag and VSVg proteins, which enhances the packaging and delivery efficiency of genetic cargo. In this specific formulation, the eExos deliver a plasmid encoding miR-135 (specifically miR-135-a-2), a microRNA identified through high-throughput screening as a potent anti-glioma agent. The mechanism involves the delivery of miR-135 into glioma stem cells (GSCs), where it acts to inhibit cell proliferation and tumor growth. Research conducted by MD Anderson Cancer Center and Baylor College of Medicine has demonstrated that these engineered exosomes can effectively cross biological barriers and deliver therapeutic miRs in both in vitro and in vivo glioblastoma models.
