Overview

eExos-miRLet7i is an experimental exosome-based gene delivery tool designed for the treatment of glioblastoma. It consists of engineered exosomes (eExos) that are modified to express retroviral Gag and VSVg proteins, which enhance the packaging efficiency of genetic cargo and facilitate entry into target cells. The therapeutic cargo in this formulation is a plasmid encoding microRNA Let-7i (miR-Let7i), which was identified through high-throughput screening as a potent anti-glioma microRNA. By delivering miR-Let7i directly to glioma stem cells, the therapy aims to inhibit tumor cell proliferation and improve survival. Preclinical studies in mouse models of glioblastoma have demonstrated that eExos-miRLet7i can significantly decrease tumor cell proliferation and, when used in combination with other microRNAs, significantly extend survival.
