EF5 is a fluorinated derivative of the 2-nitroimidazole etanidazole developed as a hypoxia marker for research and clinical imaging. It selectively binds to hypoxic cells by forming adducts with intracellular macromolecules in the absence of oxygen. This property allows EF5 to be used for detecting and quantifying tissue hypoxia in animal models and human tumors through immunohistochemistry or PET imaging when radiolabeled (e.g., 18F-EF5). The compound was developed at the University of Pennsylvania by Dr. Cameron Koch and Dr. Sydney Evans. EF5 is characterized by its lipophilicity, rapid and uniform biodistribution, high stability in humans, and ability to provide reliable detection of tumor hypoxia[1][3][5].
