Overview

**Efalizumab + topical corticosteroid** is a combination regimen used in the management of moderate-to-severe chronic plaque psoriasis. Efalizumab is a recombinant humanized monoclonal antibody targeting the CD11a subunit of LFA-1 (lymphocyte function-associated antigen-1), an immunosuppressant that disrupts T-lymphocyte adhesion and migration, thereby inhibiting the immunologic cascade involved in psoriasis[1][2][5][7]. Topical corticosteroids are synthetic glucocorticoids that act as anti-inflammatory agents, suppressing pro-inflammatory gene expression via glucocorticoid receptor activation in skin cells. The combination has been used to maximize clinical response and control flare-ups. Efalizumab was originally developed and marketed for psoriasis, but has been withdrawn globally due to severe adverse effects, including increased risk of infections and progressive multifocal leukoencephalopathy (PML)[2][3][7][8]. This regimen is no longer approved or marketed.
