Overview

Efarindodekin alfa is an **investigational, tumor-activated interleukin-12 (IL-12) immunotherapy** developed to stimulate anti-tumor immunity and reprogram the tumor microenvironment of poorly immunogenic (“cold”) tumors into an inflamed, “hot” state. The drug employs a proprietary masking technology that restricts IL-12 activation to the tumor site, aiming to enhance efficacy while minimizing systemic toxicity. It is currently being investigated in Phase 1/2 clinical trials as a monotherapy in patients with advanced solid tumors. Early clinical data demonstrate promising anti-tumor activity, durable IFNγ signaling, and a favorable tolerability profile. The program originated with Xilio Therapeutics and is being advanced in partnership with Gilead Sciences[1][3][9].
