Overview

Efbalropendekin alfa is a recombinant fusion protein consisting of a soluble interleukin-15 (IL-15) and interleukin-15 receptor alpha (IL-15Rα) heterodimer complex, fused to a half-life-extended Fc domain. It is engineered with mutations to reduce IL-15 affinity for its receptor, aiming for lower potency and therefore prolonged pharmacodynamic exposure and tolerability. Its mechanism involves stimulating the expansion and activation of T cells and natural killer (NK) cells, enhancing their cytotoxic potential against tumor cells. This agent was developed for use as an immunotherapy, notably in oncology settings such as solid tumors and multiple myeloma, and has been tested in combination with other antibodies in early-phase studies.
