Overview

Eftilagimod alpha is a recombinant fusion protein and immunotherapy drug developed for cancer treatment. It is a soluble version of the immune checkpoint molecule LAG-3, consisting of the four extracellular domains of LAG-3 fused to the Fc region of human IgG1. As an MHC class II agonist and antigen-presenting cell (APC) activator, it binds with high affinity to MHC class II molecules on dendritic cells and monocytes. This interaction leads to APC maturation and migration to lymph nodes as well as enhanced cross-presentation of antigens to T cells—particularly cytotoxic CD8+ T cells—resulting in increased anti-tumor immune responses. Eftilagimod alpha is being investigated primarily for metastatic breast cancer, non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), soft tissue sarcoma, squamous cell cancer, and COVID-19 infections. It is administered by subcutaneous injection.
