Overview

**eftozanermin alfa + venetoclax** is a combination therapy of eftozanermin alfa (formerly ABBV-621, a TRAIL receptor agonist) and venetoclax (a BCL-2 inhibitor). Eftozanermin alfa is a recombinant fusion protein that selectively binds and activates death receptors DR4 and DR5 to engage the extrinsic apoptosis pathway. Venetoclax is an orally available small molecule that inhibits the anti-apoptotic BCL-2 protein, restoring apoptosis in malignant cells via the intrinsic pathway. Together, the combination simultaneously activates extrinsic and intrinsic apoptotic pathways, which may enhance anti-leukemic efficacy. The combination has been investigated primarily in relapsed/refractory acute myeloid leukemia (AML) and diffuse large B-cell lymphoma (DLBCL)[5][1][7].
