EG-007 + lenvatinib + pembrolizumab is a combination of three drugs being studied in a global, pivotal phase III clinical trial as a first-line treatment for advanced endometrial cancer. EG-007 (developed by Evergreen Pharmaceutical/Evergreen Therapeutics) is an injectable drug which enhances anti-tumor immune response by increasing CD8+ T-cell infiltration, upregulating interferon-gamma (IFN-γ) signaling pathways, and modulating the tumor microenvironment to convert immunologically "cold" tumors to "hot." Lenvatinib is a multi-kinase inhibitor targeting primarily angiogenesis-associated receptor tyrosine kinases (including VEGFR and FGFR), and pembrolizumab is an anti-PD-1 monoclonal antibody that promotes T-cell mediated anti-tumor activity by blocking PD-1-mediated immune evasion. The triple combination is designed to synergistically target tumor growth via modulation of the immune microenvironment and blockade of immune checkpoints, with the aim of achieving improved response rates over standard treatments in advanced endometrial cancer[1][2][4][7].
