Overview

Egaptivon pegol is a PEGylated DNA aptamer that acts as an antagonist of the A1 domain of von Willebrand factor (vWF), thereby inhibiting its interaction with the platelet glycoprotein Ib (GPIb) receptor. This mechanism blocks vWF-mediated platelet adhesion, activation, and aggregation under high shear conditions, such as those found in coronary arterial stenosis and plaque rupture. Egaptivon pegol was developed for antithrombotic therapy and investigated primarily for thrombotic thrombocytopenic purpura (TTP) and acute coronary syndromes. It is a second-generation aptamer with enhanced properties due to PEGylation[1][2][7].
