Overview

EGEN-2784 is a genetically engineered porcine (pig) kidney developed as a donor organ for xenotransplantation into human patients with end-stage renal disease or kidney failure. It is the lead candidate from eGenesis and features three classes of genetic modifications to improve compatibility and safety in human recipients: (1) knockout of three genes involved in the synthesis of glycan antigens that trigger hyperacute immune rejection, (2) insertion of seven human transgenes to regulate immune response, reduce inflammation, improve coagulation compatibility, and modulate complement activation pathways, and (3) inactivation of endogenous retroviruses within the porcine genome to enhance safety. The product aims to address the critical shortage of transplantable kidneys by providing an alternative source through cross-species transplantation. The first successful transplants using EGEN-2784 were performed at Massachusetts General Hospital under FDA-authorized Expanded Access protocols[2][3][5][7].
