**EGF-CRM197** is an investigational recombinant therapeutic cancer vaccine developed by **Shanghai Humantech Biotechnology** for advanced solid tumors, especially tumors that depend on the **EGF EGFR signaling axis** such as non-small cell lung cancer. The product is a **conjugate vaccine** in which **human epidermal growth factor** is linked to **CRM197**, a non-toxic mutant diphtheria toxin carrier protein, and is administered as an emulsion with **Montanide ISA 51 VG** adjuvant. Its mechanism is to induce high titers of **neutralizing anti-EGF antibodies**, thereby depleting circulating EGF, reducing activation of **epidermal growth factor receptor**, and suppressing downstream tumor-promoting signaling. In a first-in-human phase 1 study in patients with advanced solid tumors, subcutaneous EGF-CRM197 showed acceptable tolerability, mainly grade 1 to 2 injection-site reactions, and generated robust anti-EGF antibody responses with marked serum EGF reduction; development has been reported in advanced solid tumors with a strong emphasis on lung cancer.
