An EGF neutralizing antibody is an antibody designed to bind specifically to Epidermal Growth Factor (EGF) and block its interaction with the Epidermal Growth Factor Receptor (EGFR). By sequestering circulating or local EGF, these antibodies inhibit downstream EGFR-mediated signaling pathways, such as the MAPK and PI3K/Akt pathways, which are critical drivers of cell proliferation, survival, and angiogenesis in various cancers. While research-grade EGF-neutralizing antibodies are widely used as laboratory tools to study growth factor signaling and tumor microenvironment interactions, therapeutic strategies in the clinic more commonly target the receptor itself (using anti-EGFR monoclonal antibodies like cetuximab) or utilize active immunotherapies (such as CIMAvax-EGF) to induce the patient's own immune system to produce neutralizing anti-EGF antibodies.
