EGFR CAR T refers to T cells engineered to express a chimeric antigen receptor (CAR) targeting the epidermal growth factor receptor (EGFR), a protein frequently overexpressed in various solid tumors, including glioblastoma and lung cancer. The specific research from the University of Pennsylvania describes an armored version of this therapy designed to overcome the immunosuppressive tumor microenvironment (TME). This construct incorporates a TGFβRII ectodomain fused to a switch receptor (such as CD40, OX40, or CD27), which converts inhibitory TGFβ signaling into immune-activating costimulatory signals. This dual-function platform aims to enhance T-cell survival, proliferation, and cytotoxicity while counteracting TGFβ-driven immune suppression in glioblastoma.
