EGFR-CD3 is a bispecific T-cell engager (BiTE) or bispecific antibody designed to redirect T-cell cytotoxicity against tumors expressing the Epidermal Growth Factor Receptor (EGFR). By simultaneously binding to EGFR on the surface of cancer cells and the CD3 subunit of the T-cell receptor (TCR) complex on T-lymphocytes, the molecule facilitates the formation of an immunological synapse. This interaction triggers T-cell activation, proliferation, and the release of perforins and granzymes, leading to the targeted lysis of EGFR-positive malignant cells. EGFR is a well-validated oncogenic driver overexpressed in numerous solid malignancies, including non-small cell lung cancer (NSCLC), colorectal cancer, and head and neck squamous cell carcinoma (HNSCC). Preclinical studies, such as those conducted by Revitope Oncology using humanized mouse models, have demonstrated potent anti-tumor activity, with some candidates achieving complete tumor regression in breast cancer models and maintaining durable responses after dosing cessation.
