EGFR-IL12-CART is an investigational chimeric antigen receptor T cell (CAR-T) therapy engineered to target the epidermal growth factor receptor (EGFR) and express interleukin-12 (IL-12). The therapy involves autologous T cells that are genetically modified to express a CAR specific for EGFR, commonly overexpressed in various solid tumors, and to secrete IL-12 upon activation. The secreted IL-12 enhances antitumor immune responses by promoting interferon-gamma production, activating natural killer cells, and inducing cytotoxic T cell activity. This dual mechanism aims to improve the efficacy of CAR-T therapy against solid tumors such as metastatic colorectal cancer. The drug is being developed primarily by Pregene ShenZhen Biotechnology and Shenzhen Puruijin Biological Pharmaceutical Co., Ltd., with clinical evaluation in China[2][3][4].
