EGFR-KIR CAR T is a preclinical, EGFR-targeted multi-chain chimeric antigen receptor (CAR) T cell therapy developed through a collaboration between Verismo Therapeutics and the University of Pennsylvania. Unlike conventional CAR T cells that utilize the CD3-ζ signaling pathway, this therapy employs a KIR-based signaling architecture. It combines a single-chain variable fragment (scFv) targeting EGFR variants with the KIR2DS2 transmembrane and intracellular domains and the DAP12 signaling adaptor. This "split-signal" system is designed to mimic natural killer (NK) cell activation, potentially reducing T cell exhaustion and improving persistence in the immunosuppressive environment of solid tumors like glioblastoma. Preclinical data presented at the AACR 2026 meeting demonstrated superior tumor regression and survival in murine models compared to traditional CD3-based CAR T cells.
