Overview

EGFR t-haNK is an allogeneic, off-the-shelf natural killer (NK) cell therapy developed by ImmunityBio (formerly NantKwest). It is derived from the NK-92 cell line and engineered to express a chimeric antigen receptor (CAR) targeting the epidermal growth factor receptor (EGFR), a high-affinity CD16 (V158) receptor to enhance antibody-dependent cellular cytotoxicity (ADCC), and endogenous IL-2 to promote cell survival and proliferation without the need for exogenous cytokine support. The therapy is designed to treat various EGFR-expressing solid tumors, including triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), and squamous cell carcinoma of the head and neck (SCCHN). It is frequently evaluated in combination regimens with other immunotherapies, such as the IL-15 superagonist N-803 and checkpoint inhibitors, to maximize anti-tumor efficacy.
