EGFR-TALEN is an experimental genome-editing agent designed to specifically target and disrupt the Epidermal Growth Factor Receptor (EGFR) gene. It utilizes Transcription Activator-Like Effector Nucleases (TALENs), which are engineered proteins that bind to specific DNA sequences and create double-strand breaks. These breaks are typically repaired by the cell's error-prone non-homologous end joining (NHEJ) pathway, leading to insertions or deletions that knock out the gene. In preclinical studies, EGFR-TALEN has been used to permanently silence EGFR expression in various cancer cell lines, including lung and colorectal cancer, to investigate the role of EGFR in tumor growth, invasion, and resistance to tyrosine kinase inhibitors (TKIs) or monoclonal antibodies.
