EGFR-TRAB is an experimental bispecific T-cell redirecting antibody (TRAB) developed by Chugai Pharmaceutical. It is engineered to simultaneously target the Epidermal Growth Factor Receptor (EGFR) on tumor cells and the CD3 epsilon subunit on T cells. Utilizing CrossMab technology and "knobs into holes" engineering, the molecule incorporates the complementarity-determining regions (CDRs) of cetuximab and the anti-CD3 arm of ERY974. The mechanism of action involves the formation of an immunological synapse between T cells and EGFR-expressing cancer cells, leading to T-cell activation and subsequent T-cell-dependent cellular cytotoxicity (TDCC). Preclinical research has demonstrated that EGFR-TRAB exhibits enhanced antitumor efficacy when combined with chemotherapeutic agents like paclitaxel or capecitabine, particularly in non-inflamed tumor microenvironments.
