Overview

EGFR806 CAR T cells are an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target a specific tumor-restricted epitope of the epidermal growth factor receptor (EGFR). The therapy utilizes a single-chain variable fragment (scFv) derived from the mAb806 antibody, which selectively binds to a conformationally exposed epitope (the 806 epitope) that is accessible on truncated (EGFRvIII) or overexpressed/activated wild-type EGFR, but not on the wild-type EGFR found in normal tissues. The CAR construct incorporates a 4-1BB (CD137) costimulatory domain and a CD3ζ signaling domain. Primarily developed for central nervous system (CNS) malignancies and solid tumors like triple-negative breast cancer, these cells are often delivered via locoregional routes (e.g., intracerebroventricular) to maximize local efficacy and minimize systemic toxicity.
