EGFRvIII-directed IL-10 CAR-T (also referred to as Meta 10C-based IL-10 CAR-T) is a preclinical-stage chimeric antigen receptor T-cell (CAR-T) therapy developed by Leman Bio. The therapy utilizes the proprietary 'Meta 10C' metabolic-armoring platform, which involves engineering CAR-T cells to express the immunomodulatory cytokine IL-10 alongside a CAR targeting the tumor-specific antigen EGFRvIII (epidermal growth factor receptor variant III). This approach is designed to overcome the immunosuppressive tumor microenvironment by enhancing the metabolic fitness, persistence, and anti-tumor activity of the T cells. Preclinical studies in the 4T1-EGFRvIII syngeneic mouse model of breast cancer demonstrated a 100% cure rate, suggesting potential efficacy against EGFRvIII-positive solid tumors.
