The EGFRvIII mRNA LNP vaccine is an experimental cancer immunotherapy developed by Providence Therapeutics in collaboration with the University of Calgary. It is designed to treat glioblastoma (GBM) by targeting the human epidermal growth factor receptor variant III (EGFRvIII), a tumor-specific oncogenic mutation frequently expressed in GBM but absent in normal tissues. The vaccine utilizes a lipid nanoparticle (LNP) platform to deliver messenger RNA (mRNA) encoding the human EGFRvIII neoantigen. Upon administration, the vaccine stimulates the immune system to recognize and attack EGFRvIII-expressing tumor cells. Preclinical studies presented at SNO 2025 demonstrated that the vaccine re-programs the tumor microenvironment (TME), leading to an enrichment of pro-inflammatory, antigen-processing macrophages and activated cytotoxic T-cells, which resulted in significant tumor regression and complete clearance in syngeneic mouse models.
