EGFRvIII SIR-T is a preclinical cell therapy candidate developed by Angeles Therapeutics, utilizing their proprietary Synthetic Immune Receptor T-cell (SIR-T) platform. This platform aims to improve upon conventional CAR-T cell therapies by engineering T-cells with synthetic receptors that enhance activation and durability in the tumor microenvironment. The therapy specifically targets the Epidermal Growth Factor Receptor variant III (EGFRvIII), a tumor-specific neoantigen resulting from a specific genomic deletion that is frequently expressed in glioblastoma multiforme (GBM) and other solid tumors, but absent in healthy tissues. This high degree of tumor specificity is intended to reduce off-tumor toxicities while providing a potent immune response against malignant cells.
