EGFRvIII-specific CAR-T cells (Chembrain) is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by Chembrain LTD for the treatment of glioblastoma, specifically targeting patients with leptomeningeal disease. The therapy involves genetically modifying a patient's own T cells to express a CAR that specifically recognizes the Epidermal Growth Factor Receptor variant III (EGFRvIII), a tumor-specific mutation frequently found in glioblastoma but absent in normal tissues. This specific construct is hinge-optimized and utilizes a second-generation CAR architecture incorporating a CD3ζ stimulatory domain and a 4-1BB (CD137) co-stimulatory domain to enhance T-cell activation, persistence, and anti-tumor efficacy. The cells are administered via intracerebroventricular (ICV) injection to facilitate direct access to the central nervous system and bypass the blood-brain barrier.
