EGFRxEGFR T-BsAb is a bispecific T cell-engaging antibody (T-BsAb) specifically engineered to bind two different epitopes of the epidermal growth factor receptor (EGFR) on tumor cells. By engaging EGFR in a bivalent manner and simultaneously binding to CD3 on T cells, it functions as a T cell engager, redirecting T cells to tumor cells overexpressing EGFR. This redirection promotes T cell activation and cytotoxicity against EGFR-expressing tumor cells, particularly in cancers such as pancreatic ductal adenocarcinoma (PDAC). In preclinical models, EGFRxEGFR T-BsAb displayed potent T cell-mediated cytotoxicity and tumor control, though efficacy outcomes may be lower compared to heterodimeric EGFRxHER2 T-BsAbs in some settings. Its mechanism is distinctive from conventional anti-EGFR monoclonal antibodies by promoting tumor cell killing through direct T cell engagement and cytotoxic cytokine release[2].
