EGL-001 is an investigational fusion protein drug developed for the treatment of advanced and metastatic solid tumors. It is a humanized antibody-based immunocytokine designed to selectively target and deplete regulatory T cells (Tregs) within the tumor microenvironment. EGL-001 acts as a dual antagonist, blocking both Cytotoxic T-Lymphocyte Antigen 4 (CTLA-4) and CD25 (the alpha chain of the interleukin-2 receptor), leading to inhibition of high-affinity IL-2 signaling in Tregs, internalization and downregulation of CD25, and subsequent depletion of tumor-associated Tregs. This mechanism enhances anti-tumor immune responses by increasing activation of CD8+ effector T cells while minimizing peripheral toxicities due to preferential retention in tumors. EGL-001 is being evaluated as both monotherapy and in combination with anti-PD(L)-1 checkpoint inhibitors in ongoing Phase 1/2 clinical trials for patients with recurrent or metastatic solid tumors[1][2][3][4][5][6][7].
