EGL-002 is a novel anti-CCR8/IL2 mutein fusion antibody developed by Egle Therapeutics for the treatment of cancer, specifically targeting solid tumors. The drug is designed to deplete regulatory T cells (Tregs) within the tumor microenvironment by dual binding to CCR8 and CD25 on Tregs. This dual targeting results in prolonged retention of the antibody on tumor-infiltrating Tregs, avoiding rapid internalization that limits conventional CCR8 antibodies. The mechanism leads to enhanced antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP), resulting in near-complete depletion of tumor Tregs and potent anti-tumor activity in preclinical models. EGL-002 was positioned as a best-in-class anti-CCR8 agent with potential as monotherapy or in combination with immune checkpoint inhibitors[3][6][8]. It is an engineered recombinant fusion protein classified as an immunocytokine.
