Overview

LY-354740, also known as eglumetad, is a potent and selective agonist of group II metabotropic glutamate receptors (mGlu2 and mGlu3), with nanomolar EC50 values at these receptors and minimal activity at other mGlu receptor subtypes. It is an analog of glutamate that acts to inhibit excessive glutamate release in the brain, providing anxiolytic, antipsychotic, anti-addictive, and neuroprotective effects in animal models. Clinically, it has been studied for generalized anxiety disorder with efficacy comparable to benzodiazepines but with improved tolerability. LY-354740 has also shown properties in animal models relevant to nicotine withdrawal and panic disorders. It was developed by Eli Lilly for oral administration as a small molecule therapeutic[1][2][4][6][7][10].
