Overview

EHT 6706 is a novel small molecule tubulin polymerization inhibitor that acts as both a cytotoxic agent and a vascular disrupting agent (VDA). Developed by Exonhit, it binds to the colchicine binding site on tubulin, thereby inhibiting microtubule assembly. This disruption of microtubule dynamics leads to cell cycle arrest at the metaphase/anaphase transition and induces apoptosis in a wide variety of cancer types, including breast, lung, and colorectal cancers. Notably, EHT 6706 maintains potency against multi-drug resistant (MDR) cell lines and those resistant to other tubulin-targeting agents like combretastatin and vinblastine. Beyond its direct anti-proliferative effects, it also exhibits anti-angiogenic and vascular-disrupting properties by increasing vascular permeability and disrupting pre-existing tumor blood vessels.
