EI-16004 is a novel small molecule 3-benzyl-N-phenyl-1H-pyrazole-5-carboxamide derivative being investigated for its neuroprotective and anti-inflammatory properties in the treatment of Parkinson's disease. It specifically targets astrocyte-mediated neuroinflammation by inhibiting the ERK/NF-κB signaling pathway. Mechanistically, EI-16004 attenuates the phosphorylation of extracellular signal-regulated kinase (ERK), which subsequently reduces the phosphorylation of the p65 subunit of NF-κB, leading to a decrease in the production of pro-inflammatory cytokines and chemokines in astrocytes. In preclinical models of Parkinson's disease, EI-16004 has demonstrated the ability to ameliorate motor impairment, protect against dopaminergic neuronal loss, and mitigate astrocyte activation in the striatum and substantia nigra.
