EI-201 is an investigational mRNA-based immunotherapy developed by eTheRNA immunotherapies for the treatment of HPV16-positive carcinomas. It consists of mRNAs encoding the HPV16 oncoproteins E6 and E7, which are the primary biological targets, combined with eTheRNA's proprietary TriMix mRNA adjuvant. TriMix is designed to activate dendritic cells and stimulate a robust T-cell immune response against tumor cells expressing these viral antigens. The mRNA components are encapsulated in lipid nanoparticles (LNPs) to facilitate delivery to antigen-presenting cells following intravenous administration. EI-201 is currently being evaluated in Phase I/IIa clinical trials for patients with recurrent or metastatic HPV16-positive cancers, including cervical, anal, and oropharyngeal cancers.
