eIF5A plasmid DNA, specifically the pExp5A plasmid, is a gene therapy component designed to express a non-hypusinable, pro-apoptotic mutant of the eukaryotic translation initiation factor 5A (eIF5A), known as eIF5AK50R. Developed by Senesco Technologies (now Sevion Therapeutics), this plasmid is a critical part of the SNS01-T nanoparticle therapeutic. The plasmid is engineered with a B-cell-specific promoter/enhancer to ensure selective expression of the pro-apoptotic mutant in malignant B-cells. In the SNS01-T formulation, the plasmid is co-delivered with an eIF5A siRNA via polyethylenimine (PEI)-based nanoparticles. This dual mechanism induces cancer cell death by simultaneously suppressing the survival-promoting hypusinated form of endogenous eIF5A and overexpressing the pro-apoptotic eIF5AK50R variant, primarily for the treatment of multiple myeloma and other B-cell malignancies.
