Drug intelligence / Profile preview
EIK1003 + abiraterone is an investigational combination regimen being developed by Eikon Therapeutics and IMPACT Therapeutics for the treatment of advanced prostate cancer. EIK1003 (formerly IMP1734) is a highly selective, non-brain-penetrant small-molecule inhibitor of poly(ADP-ribose) polymerase 1 (PARP1) that spares PARP2, thereby potentially reducing hematological toxicities compared to non-selective PARP inhibitors. Abiraterone is an approved novel hormonal agent that acts as a potent, selective, and irreversible inhibitor of cytochrome P450 17A1 (CYP17A1), a key enzyme in androgen biosynthesis. The combination, typically administered orally once daily alongside prednisone or prednisolone, is being evaluated in the global Phase 1/2 EIK1003-001 clinical trial (NCT06253130) in patients with metastatic prostate cancer, including metastatic castration-resistant prostate cancer (mCRPC) and metastatic castration-sensitive prostate cancer (mCSPC) harboring homologous recombination repair (HRR) gene alterations.
