Overview

EIK1004 is a central nervous system (CNS)-penetrant, potent and selective PARP1 inhibitor developed for the treatment of advanced solid tumors, including those with brain metastases. It is designed to selectively inhibit PARP1 (poly(ADP-ribose) polymerase 1), an enzyme involved in DNA repair, with the goal of inducing synthetic lethality in cancer cells harboring homologous recombination repair (HRR) gene mutations. Eikon Therapeutics is developing EIK1004 as monotherapy for patients with recurrent, advanced/metastatic breast cancer, ovarian cancer, metastatic castrate-resistant prostate cancer (mCRPC), and pancreatic cancer[2][3][5]. The drug has demonstrated CNS penetration in preclinical studies and is currently being evaluated in Phase 1/2 clinical trials for safety, tolerability, pharmacokinetics/pharmacodynamics, and preliminary antitumor activity[5][6].
