Drug intelligence / Profile preview
EIK1005 + pembrolizumab is an investigational combination therapy consisting of EIK1005, a novel small-molecule inhibitor of Werner (WRN) helicase, and pembrolizumab, a marketed anti-PD-1 monoclonal antibody (Keytruda). Developed by Eikon Therapeutics in collaboration with Merck & Co., this combination is designed to exploit synthetic lethality in microsatellite instability-high (MSI-H) or mismatch repair-deficient (dMMR) tumor cells, which depend on the WRN helicase pathway to maintain genome integrity, while simultaneously enhancing the immune system's ability to target and destroy cancer cells. The combination is being evaluated in a Phase 1/2 clinical trial (NCT07262619) for patients with advanced solid tumors, including MSI-H/dMMR tumors.
