EKA4 is a fully human monoclonal antibody (IgG4) that targets prostatic acid phosphatase (ACP3), a protein highly expressed in prostate epithelial cells and a clinically relevant antigen in prostate cancer. Developed using phage display technology from a naïve scFv library, EKA4 exhibits high affinity with a dissociation constant (KD) of 5 nM in its scFv format. It was subsequently cloned and expressed as a full-length IgG4 antibody in CHO cells. EKA4 is being investigated as a potential scaffold for the development of next-generation tumor-targeted therapeutics, particularly for patients with prostate cancer who do not benefit from PSMA-targeted therapies.
