Overview

Elacestrant + triptorelin is a combination therapy under investigation for the treatment of hormone receptor-positive (HR+), HER2-negative breast cancer in premenopausal women. Elacestrant is an oral selective estrogen receptor degrader (SERD) that binds to and degrades the estrogen receptor (ER), thereby blocking estrogen-driven tumor growth. Triptorelin is a gonadotropin-releasing hormone (GnRH) agonist that suppresses ovarian function by downregulating luteinizing hormone (LH) and follicle-stimulating hormone (FSH), leading to reduced production of endogenous estrogens. The combination aims to maximize endocrine suppression in premenopausal patients by pairing direct ER degradation with ovarian function suppression. This regimen is being evaluated in clinical trials such as the PREMIERE study for its biological effects and efficacy compared to elacestrant alone[5][7].
