Overview

Elafibranor is a small molecule drug that acts as a dual agonist of peroxisome proliferator-activated receptor alpha (PPARα) and delta (PPARδ). It is indicated for the treatment of primary biliary cholangitis (PBC), either in combination with ursodeoxycholic acid (UDCA) in adults who have had an inadequate response to UDCA, or as monotherapy in patients unable to tolerate UDCA. Elafibranor works by decreasing the production of bile acids and increasing their movement out of the liver, thereby reducing toxic bile acid accumulation. Its mechanism involves modulation of lipid and glucose metabolism, mitochondrial fatty acid metabolism, oxidative stress reduction, and improvement in bile acid homeostasis. The drug was originally developed by Genfit and later licensed to Ipsen for global development and commercialization outside Greater China; Terns Pharmaceuticals holds rights for Greater China[1][2][4][5][9].
