ELB041 is a humanized monoclonal antibody targeting the macrophage colony-stimulating factor receptor CD115 (CSF1R) on myeloid-lineage cells, developed by Elsalys Biotech after initial discovery at Transgene, for use in immuno-oncology and fibrotic indications.[1][6][12] By selectively modulating, rather than fully blocking, M-CSF/CD115 signaling, ELB041 inhibits differentiation and survival of immunosuppressive M2 tumor-associated macrophages while sparing pro-inflammatory M1 macrophages, thereby relieving myeloid-driven immunosuppression and potentially enhancing antitumor immunity.[1] Unlike other anti-CD115 antibodies, it does not prevent ligand binding but attenuates receptor activity in a way that avoids elevation of circulating M-CSF and is expected to confer a more favorable safety profile; preclinical development also includes evaluation in bleomycin- and radiation-induced lung fibrosis models, leveraging its capacity to reshape pathogenic myeloid compartments.[1][6]
