ELBPRA is an engineered live biotherapeutic product (LBP) based on the *Escherichia coli* Nissle 1917 (EcN) strain, designed for the treatment of inflammatory bowel disease (IBD). Developed by researchers at Tianjin University, ELBPRA is genetically modified to synthesize and deliver all-trans retinoic acid (ATRA) directly to the gut. The engineering involves expressing genes for β-carotene synthesis and its subsequent conversion to retinoic acid via β-carotene oxygenase and retinal dehydrogenase. To overcome solubility issues, the strain is modified to release ATRA via extracellular vesicles. In preclinical models of colitis, ELBPRA has demonstrated the ability to reduce weight loss, improve colon structure, and modulate inflammatory cytokines, specifically increasing IL-10 and decreasing TNF-α and IL-1β.
