ELD607 is a novel immunomodulator and store-operated calcium entry (SOCE) inhibitor that selectively targets the Orai1 calcium channel. By inducing the internalization of Orai1 from the plasma membrane, ELD607 suppresses intracellular calcium signaling, which is a critical regulator of fibroblast functions such as proliferation, extracellular matrix (ECM) production, and contraction. In preclinical models of idiopathic pulmonary fibrosis (IPF), ELD607 has demonstrated the ability to reverse TGF-β-induced α-smooth muscle actin (α-SMA) expression and ECM production in lung fibroblasts. Furthermore, it has shown efficacy in reducing lung damage and collagen deposition in bleomycin-exposed mice when administered via inhalation. The drug is being investigated as a potential therapeutic for chronic fibrotic lung diseases.
