ELD615

ELD615 is an inhaled peptide-based Orai1 modulator and degrader being developed by Eldec Pharmaceuticals for the treatment of chronic lung diseases, specifically non-cystic fibrosis bronchiectasis (NCFB). Derived from a sequence of the SPLUNC1 C-terminus, ELD615 binds extracellularly to Orai1 (calcium release-activated calcium channel protein 1), a calcium channel that acts as an upstream gatekeeper in the pro-inflammatory signaling cascade. This binding triggers the internalization and degradation of the channel, thereby blocking Orai1-mediated pro-inflammatory signaling. In preclinical models of bronchiectasis, ELD615 has demonstrated the ability to reduce neutrophil-driven inflammation, decrease mucus production and airway plugging, lower bacterial burden, and reverse existing airway damage.
