Overview

Elegrobart (VRDN-003) is a subcutaneous monoclonal antibody developed by Viridian Therapeutics for the treatment of thyroid eye disease (TED). It acts as an inhibitor of the insulin-like growth factor-1 receptor (IGF-1R). Engineered from its parent molecule, veligrotug, elegrobart features the same binding domain but is modified with Fc-optimization to extend its half-life, enabling less frequent dosing. It is designed for self-administration via a commercially validated autoinjector, with dosing schedules of every four or eight weeks. The drug is currently being evaluated in two global Phase 3 pivotal trials, REVEAL-1 and REVEAL-2, for active and chronic TED, respectively.
