Elemene + EGFR-TKIs is a combination therapy under investigation for the treatment of advanced non-small cell lung cancer (NSCLC) with activating mutations in the epidermal growth factor receptor (EGFR). Elemene (specifically β-elemene) is a natural sesquiterpene compound derived from the Chinese medicinal herb Rhizoma Zedoariae. It exhibits anti-tumor properties and has been shown to synergize with EGFR tyrosine kinase inhibitors (TKIs), such as gefitinib and osimertinib. The combination aims to enhance anti-cancer efficacy by inhibiting cellular proliferation and migration while overcoming or delaying resistance mechanisms associated with long-term TKI use. Elemene may also reverse TKI resistance by suppressing autophagy linked to lncRNA H19 and impairing cancer stem-like properties[2][3][8]. Clinical trials are ongoing in China to evaluate its safety and efficacy as an adjunctive therapy in patients with advanced NSCLC harboring EGFR mutations[4][8].
